Reimagining Psychedelic Therapeutics for Daily Outpatient Use: Advancing Low-Dose Psilocin Beyond the Hallucinogenic Paradigm

- Stable, precisely dosed, and potentially self-administered: L-130 is a patented salt of psilocin, the active moiety itself, not a prodrug, with greater than two-year shelf stability and consistent oral delivery. Because anxiolytic effects were observed without hallucinations, L-130 may support an outpatient, take-home treatment model rather than the supervised in-clinic dosing that psychedelic-class candidates typically require.
- First-in-man results show near-complete bioavailability with unusually low inter-subject variability: Across 10 subjects, Cmax and Tmax were highly consistent. At 2 mg psilocin equivalent, subjects reported anxiolytic effects with no hallucinations or visual distortions and no serious adverse events.
- Preclinical data support low-dose daily administration: In a 28-day rodent study, daily low-dose administration produced significantly greater effect than weekly dosing at the same dose, a departure from the intermittent high-dose paradigm typical of psilocybin-class candidates.
